What are the notable acute adverse effects for therapeutic doses of psilocybin in the treatment of depression and anxiety? [Date Sources:] MEDLINE via PubMed, Web of Science, and ClinicalTrials.gov were searched for publications available between 1966 and November 30, 2023.
In this meta-analysis of 6 randomized, double-blind clinical trials with 528 patients, headaches, nausea, anxiety, dizziness, and fluctuations in blood pressure occurred significantly more frequently with psilocybin vs comparators … these side effects were generally well tolerated by participants and faded within 24 to 48 hours. Psilocybin use was not associated with risk of paranoia and transient thought disorder … “The acute side effects are what we may expect from your traditional antidepressants because those medications work in a similar fashion to psilocybin. They both target serotonin receptors,” said Joshua Caballero, corresponding senior author of the paper and an associate professor in UGA’s College of Pharmacy. “It’s very encouraging because the studies we examined consist of just one or two doses per patient, and we’re finding that the beneficial effects of psilocybin may stay for months when treating depression.”
